Offer Description
Researcher for a project on metabolic engineering in microorganisms of industrial interest, entitled ‘Upcycling of brewers’ spent grain for the biotechnological production of green hydrogen within a circular economy framework’ (BIO-H2Verde)
The successful candidate will carry out the following tasks, amongst others:
Study of the growth conditions of selected strains of Escherichia coli and Rhodobacter capsulatus to optimise hydrogen production.
Genetic modification of Escherichia coli to obtain hydrogen-producing mutant strains. To this end, plasmid constructs will be generated to produce strains with molecular hydrogen sensors, and combinatorial libraries will be created to modulate the expression of carbon metabolism enzymes involved in biohydrogen production.
Genetic modification of Rhodobacter capsulatus to generate biohydrogen-producing mutants using random mutagenesis, as well as the creation of strains via targeted mutagenesis to eliminate negative regulators—such as the presence of ammonium—in nitrogenase expression.
Specific qualifications: Bachelor’s degree in Biotechnology, Biology or Biochemistry, and a Master’s degree in Biotechnology, Bioinformatics or a similar subject.
Experience:
Molecular biology: cloning, library design and construction, NGS sequencing, quantitative PCR for metabolic engineering in Escherichia coli and Rhodobacter capsulatus.
Analytical techniques: HPLC, GC-MS, omics techniques (RNA-seq transcriptomics, metabolomics or fluxomics). Analysis of enzymatic activity using fluorescence.
Bioinformatics, including the analysis of big data or omics data, metabolic modelling and multivariate statistics using software such as R and Python, amongst others. Computational modelling of macromolecules.
Languages: English, level B2 or above
