Position Overview
The Single Cell and Synthetic Genomics of Blood Formation laboratory at the Centre for Genomic Regulation (CRG) in Barcelona is seeking a dedicated PhD candidate with a robust background in computational biology and bioinformatics. The successful applicant will spearhead a project focused on the clonal dynamics of blood formation, particularly in the context of aging and hematological disorders.
Understanding blood formation is crucial due to its biomedical significance and inherent complexity. Our research group delves into adult hematopoiesis through the application of computational biology, single cell genomics, and genetic screening methodologies.
Humans possess a limited number of blood stem cells, approximately 100,000, most of which remain in a quiescent state. Only a select few clones are actively engaged in blood production. This is vital as distinct stem cell clones exhibit both epigenetic and functional variations; for instance, some clones are more proficient in generating T cells, while others are better suited for inflammatory responses. The decline in clonal diversity and functionality is a hallmark of blood aging. Current methodologies to investigate these phenomena are lacking. We require innovative single cell techniques that can simultaneously capture a cell's functional capabilities and its clonal identity. Recently, we have developed a novel method, EPI-Clone, which allows for the acquisition of clonal, functional, and molecular data by analyzing the DNA methylome at single cell resolution across thousands of cells. The new PhD candidate will build upon this innovative approach to explore clonal dynamics in aging and hematological malignancies.
Key Responsibilities
• Collaborate with an experienced technician and clinical partners to advance this project.
• Analyze data generated from a state-of-the-art single cell DNA methylome technique.
• Work alongside fellow bioinformaticians to enhance the EPI-Clone method and related clonal tracking techniques.
• Engage closely with both experimental and computational scientists within the team.
About Our Team
Our interdisciplinary group is dedicated to investigating the biology of hematopoietic stem cells through a combination of single cell genomics, genetic screening, and computational approaches. We aim to develop innovative genomic and bioinformatics tools to address longstanding questions in the field, focusing on two primary research areas: a) Gene regulation in healthy hematopoiesis and b) Clonal functionality in aging and disease. Established in January 2020, our lab currently comprises six members, including two PhD candidates, two bridging postdoctoral researchers, one postdoctoral researcher, and two senior technicians. Our research is supported by funding from various esteemed organizations.
Candidate Profile
Essential Qualifications
• A Master’s degree in a relevant discipline (e.g., computational biology, genomics, biomedicine, bioinformatics).
• A keen interest in single cell genomics technologies and stem cell biology.
• Research experience in computational biology (minimum of 6 months on a single project).
• Strong academic performance in quantitative subjects (mathematics, physics, etc.).
• Proficient programming skills in R or Python.
Preferred Qualifications
• Additional training in experimental techniques (e.g., cloning, library preparation, qPCR, etc.).
• A solid foundation in machine learning and statistical analysis.
Language Proficiency
• Proficiency in English is required.
Technical Skills
• Data analysis expertise.
• Proficiency in R and Python programming.
