JOB OVERVIEW:

The IGTP is seeking to recruit a predoctoral researcher to join the Innate Immunity Group.

The advancement of immune checkpoint inhibitors has greatly enhanced the prognosis for individuals with advanced cancers. Nevertheless, a significant number of patients either do not respond to these treatments or ultimately develop resistance. The integration of immunotherapies that target various immunosuppressive pathways may improve treatment effectiveness or overcome resistance.

Tumor-associated macrophages (TAMs) are influenced by the tumor microenvironment to adopt a tolerogenic phenotype, which facilitates tumor growth. These cells represent the primary immune infiltrate within tumors, making them a highly attractive therapeutic target in the field of immuno-oncology.

Our objective is to further develop a pioneering immunotherapy aimed at TAMs. We have identified a crucial checkpoint in the formation of tolerogenic macrophages, which holds significant potential for numerous solid tumors. Our research involves studies on TAM biology, encompassing clinical observations, fundamental research, and mathematical modeling, to aid in the creation of a new monoclonal antibody-based therapy capable of reprogramming TAMs from a tolerogenic state to a pro-inflammatory one. These investigations will be complemented by preclinical studies in murine models; preliminary findings indicate that our therapy modifies the intratumoral microenvironment, alters the immune cell population profile (including TAM phenotype), reduces blood vessel density, and enhances the inflammatory environment, ultimately leading to decreased tumor progression.

The aim of this PhD thesis is to deepen our understanding of TAM biology, contribute to the advancement of our immunotherapy by clarifying its complete mechanism of action and potential applications, and ultimately propel the development of a novel cancer treatment. A unique aspect of this position is the opportunity for the PhD candidate to collaborate closely with TAM Therapeutics, our spin-off focused on the regulatory, clinical, and commercial progression of this research. This collaborative relationship will provide the candidate with invaluable insights into the translational facets of scientific discovery, bridging the divide between academia and industry. The candidate will not only gain expertise in cutting-edge research but also acquire essential knowledge and skills for the practical application of findings in preclinical and clinical contexts, significantly enhancing their academic and professional growth.

QUALIFICATIONS AND EXPERIENCE:

Required:

  • Candidates must possess a BSc and MSc degree in biological or biomedical sciences at the time of recruitment, enabling them to enroll in an official PhD program. Verification of the required educational level will be conducted by the host university during the admission process.
  • BSc qualification must be greater than 8/10.
  • Applicants must exhibit an advanced proficiency in the English language (both spoken and written).
  • Willingness to conduct preclinical studies in murine models under appropriate guidance and supervision.

MAIN RESPONSIBILITIES:

Experimental Work:

  • Conduct in vitro functional and phenotypic analyses of macrophages and immune cells in the context of cancer using primary cells and cell lines.
  • Participate in the execution of preclinical cancer models in murine systems and sample analysis.
  • Investigate biomarker expression in human samples using techniques such as immunohistochemistry, immunofluorescence, or RT-PCR-Sequencing.

Additional Responsibilities:

  • Maintain a thorough understanding of the research field through literature review.
  • Analyze and interpret experimental data, contributing to manuscript preparation.
  • Actively engage in laboratory meetings, IGTP seminars, and national/international conferences.
